Prokaryotic Expression of Brucella BAB Gene and Establishment of iELISA

In this study,the genome of Brucella Rev.1 strain was used as a template to amplify the BAB gene sequence,clone it into the prokaryotic expression vector pET-30a(+),obtain the recombinant plasmid pET-30a-BAB,and perform prokaryotic expression on the recombinant plasmid.The indirect ELISA method was constructed by using the expression products detected by Western blotting.The results showed that the BAB gene was successfully cloned and expressed in this experiment,and the purified expression product was analyzed by SDS-PAGE.This study obtained a relatively pure recombinant BAB protein;Western blotting test showed that the expressed protein could react specifically with Brucella sheep positive serum and had good reactogenicity;Using the recombinant BAB protein as the coating antigen,an indirect ELISA method for detecting BAB antibodies was established and optimized.The best determined coating conditions were as follows BAB protein coating amount was 0.25 μg/mL,serum dilution was 1:400;blocking solution was 3% pig-derived gelatin;secondary antibody dilution was 1:6 000;color development time was 10 min.The established method was used to detect 40 clinical sheep serums,and the cut-off value was calculated to be 0.607.That was,when the serum tested had P/N ≥ 1.5 and D_{450 nm} ≥ 0.607,it was judged as positive,when D_{450 nm} ≤ 0.561,it was judged as negative,and when 0.607<D_{450 nm}<0.561,it was judged as a suspect value,and retest was required.Compared with the Huhong plate test and the test tube agglutination test,the positive coincidence rate was 100%,the negative coincidence rate was 71.88%,and the total coincidence rate was 77.5%.     

大豆不同亲本正反交后代开花期叶片叶绿素含量遗传研究

2002年,对大豆不同亲本正、反交F3、BC1F2开花期叶片叶绿素含量遗传进行了初步比较研究,结果表明,各组合F3开花期叶片叶绿素含量平均值偏向于母本,广义遗传力的大小与母本值关系密切,母本值高的组合F3广义遗传力明显高于母本值低的组合,各组合正、反交F3开花期叶片叶绿素含量的变异度与母本值也相关,母本值高的组合F3变异度大于母本值低的组合;BC1F2开花期叶片叶绿素含量的遗传表现与组合的父母本和回交亲本的开花期叶片叶绿素含量直接相关.

农村小额信贷的社会影响评价分析——以非介入性研究《人民日报》文献为例

采用非介入性研究方法,检索、查阅了2000年至2006年《人民日报》发表的关于农村小额信贷的文章,对其在当前中国农村社会的影响进行了归纳、分析和评价,并提出了今后发展的建议。     

The Correlation Between Polymorphisms of Exon1 and Exon4 of DRB1 Gene and Brucellosis in Kazakh Sheep

The purpose of this experiment was to study the correlation between exon 1 and 4 polymorphisms of DRB1 gene and brucellosis in Kazakh sheep.Using RBPT serological tests to try sheep serum,reference in GenBank sheep MHC Class Ⅱ area DRB1 gene sequences (Accession No.:NC_040271.1),the exon 1 and 4 pieces designed primers,using PCR-SSCP and DNA sequencing technology to 230 Kazak sheep DRB1 gene polymorphism detection,analyze its polymorphism loci and the relationship between the Kazak sheep Brucella susceptibility.The results showed that 66 Kazakh sheep were positive for Brucella in RBPT test,and the positive detection rate was 28.7%.There was one SNP locus (F1-G22A) in exon 1 fragment,and sequencing determined two genotypes (GG and GA),the dominant allele and genotype were G and GG respectively,and the susceptibility genotype of the polymorphisms of F1-G22A was GA.Chi-square test showed that there was no significant correlation between the polymorphisms of DRB1 gene F1-G22A and Brucella susceptibility in Kazakh sheep (P>0.05).According to the analysis of bioinformatics online software,the F1-G22A polymorphic sites lead to the change of RNA secondary structure and the decrease of minimum free energy,and lead to the change of protein secondary structure.No SNPs were found in DRB1 exon 4 fragment.Therefore,there might be a certain correlation between the polymorphisms of DRB1 gene F1-G22A and Brucella susceptibility in Kazakh sheep.     

北京地区独本菊品种资源及花型和花色分布研究

经过多年的实地调查和记录,统计到北京地区的常见独本菊品种1 001个;根据<中华人民共和国菊花品种DUS测试指南>的要求,对这些品种进行测试分析.根据中国菊花研究会制定的品种分类标准进行品种数量统计结果表明:各种瓣型中,平瓣类(42%)和管瓣类(33%)品种数量最多,桂瓣类(2%)较为稀少;在各种花型中,叠球型205个、翻卷型97个和卷散型86个,而管球型和全桂型仅各有1个品种,极为珍贵;花色以白色、黄色居多,红色、绿色和橙色等花色在菊花品种中较为稀少,仅有翻卷型、卷散型、钩环型和管盘型花色齐全.目前生产中仍然存在大量品种名称混杂的现象,现行的菊花品种分类系统无法将这些品种区分开来.根据上述调查结果,提出需要对珍稀花型和花色品种加以特殊保护,对现行分类体系进行修正的建议,并阐述了未来引种和育种可能的发展方向,以期为菊花品种资源的可持续开发和利用提供参考.     

Effect of atorvastatin on neointimal hyperplasia of venous autografts in rats

AIM: To investigate the effect of atorvastatin on neointimal hyperplasia of autogenous vein graft in rats. METHODS: The model of autogenous vein graft was prepared by transplanting the external jugular vein into aorta in Wistar rats. The rats were divided into three groups: sham operation group, graft control group and graft experimental group. From three days after transplantation, the rats of autograft experimental group were treated by atorvastatin at a dosage of 5 mg·kg-1·d-1. Four weeks after treatment, venous autografts were removed at autopsy and cut into 4 μm sections. Histopathological examination was carried out to analysis the neointimal hyperplasia of grafted veins. Immunohistochemical staining was conducted to evaluate SMα-actin and PCNA expression of neointimal cells in venous autografts. RESULTS: In venous autograft control and experimental groups, SMα-actin-positive smooth muscle cells were proliferated and accumulated excessively in venous autografts, which resulted in significant neointimal formation and vascular lumen narrowing. Neointima quantitative assay revealed that the neointimal hyperplasia of venous autografts was suppressed obviously in graft experimental group, and its neointimal area and NIA/MA ratio of venous autografts were significantly lower than those in graft control group (P<0.01). Immunohistochemical assay indicated that the PCNA labeling index of neointimal cells was significantly lower in graft experimental group than that in graft control group (P<0.01). CONCLUSION: Atorvastatin significantly inhibits the proliferation of neointimal smooth muscle cells and the development of neointimal hyperplasia of venous autografts in rats. Atorvastatin is a powerful inhibitor of restenosis after vascular reconstructive operation with a potential for therapeutic use.
